Beyond Viability: Advancing CHO Cell Culture Process Strategies to Modulate Host Cell Protein Levels While purification strategies usually offer substantial HCP clearance, certain problematic HCPs, particularly lipases, pose significant challenges. This study investigates the accumulation of various problematic HCPs in CHO cell culture using transcriptomics, revealing correlations between cell culture parameters and HCP activity. Contrary to conventional assumptions, viability alone does not reliably predict HCP levels, with factors such as clone selection, host cell line choice, and media feed type significantly influencing HCP accumulation. Leveraging transcriptomics-based approaches, we demonstrate the potential of upstream process control strategies to mitigate HCP presence and improve biologic product quality. Accelerating Cell Culture Media Development Using Bayesian Optimization-Based Iterative Experimental Design Here, we have applied a Bayesian Optimization-based iterative framework for experimental design to accelerate cell culture media development for two applications. First, we show this approach yields new compositions of media with cytokine supplementation to maintain the viability and distribution of PBMCs in culture. Second, we applied this framework to optimize the production of three recombinant proteins in K.phaffii cultivations. For both applications, we identified conditions with improved outcomes compared to the initial standard media using 3 to 30 times fewer experiments than other methods such as the Design of Experiments. Subsequently, we also demonstrated the extensibility of our approach to efficiently account for additional design factors through transfer learning. https://lnkd.in/g9c_7ZxK #aspenalert #biotech #bioprocess
