In addition to identifying chemical starting points for early drug discovery processes, DNA-encoded library (DEL) technology has now expanded to new applications, including the discovery of bifunctional affinity ligands historically considered difficult to develop. At this year’s Gordon Research conference, WuXi AppTec presented a poster describing how DEL can enable the rapid discovery of novel affinity ligands, such as tracers for biochemical and cellular assays, and building a ‘universal’ chimeric antigen receptor (CAR) to enhance the control and safety profile of CAR-based immunotherapies. Apoptosis is defined as programmed cellular death. Currently available methods do not allow to measure cell-specific apoptosis in high-throughput. Biocytometry represents a promising method for the single-cell analysis of cellular immunophenotypes. It utilises bioparticles expressing antibody mimetics that recognise antigen markers and in response produce a luminescent reporter signal. This paper provides valuable insights into enhancing EV-based therapeutic delivery systems and opens up new possibilities for customized cargo delivery using engineered CHO cells. Highlights:** - CHO cells, commonly used for recombinant therapeutic protein production, can also serve as EV producer cells. EVs are promising for delivering bioactive cargo. - Genetic constructs and split green fluorescent protein technology were used to engineer CHO cells for custom EV production. - The custom EV approach achieved significantly higher cargo loading compared to passive strategies. - Successful delivery of engineered cargo was observed in recipient cells.
