Bioprocess Feeding Optimization Through In-Silico Dynamic Experiments and Hybrid Digital Models - A Proof of Concept In this work we suggest a methodology to perform the feeding schedule optimization of mammalian cell cultures by virtualizing part of the experimental campaign on a hybrid digital model of the process, which accelerates the experimentation and reduces the experimental burden. The proposed methodology couples Design of Dynamic Experiments (DoDE) with a hybrid semi-parametric digital model. In particular, DoDE is used to design optimal experiments with time-varying factors’ profiles whose experimental data are then utilized to train the hybrid model which is able to identify the optimal time profiles of glucose and glutamine maximizing the antibody titer in the culture despite the limited number of experiments performed on the process. https://lnkd.in/ghabD9Az #aspenalert #biotech #bioprocess

I’m excited about Illumina’s recent acquisition of Fluent Biosciences and their innovative, largely instrument-free, emulsion-based single-cell sequencing method, PIP-seq. I have great confidence in Illumina’s R&D team to elevate PIP-seq, potentially making it a strong competitor against established methods from 10x Genomics, Parse, and Scale. However, there are two key areas that I think need further attention: 1. 𝗘𝗻𝗵𝗮𝗻𝗰𝗲𝗱 𝗦𝗮𝗺𝗽𝗹𝗲 𝗙𝗶𝘅𝗮𝘁𝗶𝗼𝗻 𝗦𝘂𝗽𝗽𝗼𝗿𝘁: The current documentation for the PIP-seq DSP/Methanol fixation protocol is quite basic and needs improvement. Storing and sequencing millions of cells from multiple different samples is still an underestimated logistical challenge. 2. 𝗛𝗶𝗴𝗵-𝗧𝗵𝗿𝗼𝘂𝗴𝗵𝗽𝘂𝘁 𝗦𝗮𝗺𝗽𝗹𝗲 𝗣𝗿𝗼𝗰𝗲𝘀𝘀𝗶𝗻𝗴: As other single-cell methods have introduced automated workflows to minimize hands-on time, it’s crucial to focus on efficiency.
