Fasting has been found to be one of the most potent triggers for autophagy because when we fast, our body shifts from using glucose (from food) as its primary energy source to burning fat for energy. This shift, combined with the decrease in nutrient availability, signals cells to initiate autophagy as a way to maintain energy and remove any dysfunctional components that might have built up. During fasting, insulin levels drop because there's less food (particularly carbs) being consumed. Low insulin is a key trigger for autophagy. Fasting also increases levels of human growth hormone (HGH), which is also associated with stimulating autophagy and promoting tissue repair. Fasting, like a survival mechanism, induces mild stress on cells, which activates autophagy to help protect against damage and inflammation. Most studies suggest that autophagy begins to kick in after about 16-24 hours of fasting, with maximum effects seen around the 48-hour mark.

In a fascinating experiment, researchers discovered that exposing old mice to young blood could rejuvenate their aging hearts. By surgically connecting young and old mice in a process called parabiosis, they found that the enlarged hearts of old mice shrank back to a youthful size in just 4 weeks. The secret? A protein called GDF11, abundant in young blood but scarce in older animals. Further studies revealed that restoring GDF11 levels in old mice reversed age-related heart enlargement, showing the potential of this protein for future therapies. Our review explores the diverse roles of GPCR68 in both normal physiology and disease conditions, providing insights that could open new research avenues.
