    A recent study published in Cell Reports reveals an important connection between RNA localization and how cells migrate. Leveraging MERFISH technology, researchers from Yale University, National Cancer Institute (NCI), University of Colorado Boulder and Memorial Sloan Kettering Cancer Center, discovered that hundreds of mRNAs localize to focal adhesions (FAs). These FA-associated RNAs form complexes with G3BP1, a key RNA-binding protein, which regulates FA protein dynamics and ultimately, cell migration. This research adds a new layer to our understanding of gene regulation and could inform future studies in wound healing, cancer metastasis, and mechanobiology. In this study, MERFISH enabled: ➡️High-Resolution Spatial Mapping: Enabled precise visualization of FA-mRNAs within cells, confirming their localization at FAs. ➡️Transcriptome-Wide Insights: Identified and mapped hundreds of FA-enriched mRNAs, revealing their association with G3BP1 and FA proteins. ➡️Mechanistic Validation: Allowed researchers to study spatial gene expression changes in real time, demonstrating how FA-mRNAs interact with FA proteins. ➡️Cell Migration Research: Provided crucial evidence linking RNA localization, FA function, and cell motility, deepening our understanding of RNA-protein interactions in cytoskeletal dynamics.     New Insights into Aging Microglia with Spatial Transcriptomics ✨ A new pre-print study from researchers at Stanford University reveals how aging alters the function, morphology, and mRNA distribution of microglia. The study utilized MERFISH to analyze single-cell gene expression and morphology. By providing spatial context to gene expression data, the MERSCOPE® Platform enabled a deeper understanding of how microglia change with age. Key Findings: ➡️Subcellular mRNA localization: Aging disrupts transcript distribution within microglial processes, impacting functions like synaptic maintenance and migration. ➡️Morphological heterogeneity: Detected diverse microglial shapes within transcriptomically defined populations, including disease-associated microglia with unexpected morphologies. ➡️Functional shift with age: Aging shifts microglial functions from synaptic maintenance in younger brains to migration and catabolic pathways in older ones.     Spatial Spotlight #7 March is National Kidney Month, making it the perfect time to spotlight the complexity of the human kidney cortex through the lens of spatial transcriptomics. This MERSCOPE™ data image showcases the intricate filtering units called glomeruli and the different epithelial cells of the tubules, captured from an FFPE human kidney cortex sample.
