Study Highlights Claudin-5 in Multiple Sclerosis Using in vivo-jetPEI for Gene Silencing Researchers at the University of South Florida and Boise State University have uncovered a key player in multiple sclerosis (MS) progression through their work on experimental autoimmune encephalomyelitis (EAE), a model for MS. The protein non-muscle myosin light chain kinase (nmMLCK) was found to weaken the brain's protective barrier, the blood-brain barrier (BBB), during inflammation. Moreover, the study highlights the importance of another protein, claudin-5, in maintaining the BBB's integrity. Using in vivo-jetPEI for gene silencing, they found that reducing claudin-5 levels worsened the disease, emphasizing its protective role. These insights pave the way for new treatments targeting nmMLCK and enhancing claudin-5 to protect the BBB, potentially slowing MS progression.
