Why Choose #mIHC? ➡️ Preserves Morphology: With chromogenic dyes, mIHC offers unmatched clarity for evaluating tissue structures like epithelial architecture and stroma. ➡️ Visual Co-Localization: Chromogenic dyes, particularly Roche’s translucent purple, yellow, and teal, enable the visualization of multiple markers while preserving tissue morphology. These dyes blend to reveal co-localization through distinct new colors: 🎨 Purple + Yellow: Fiery red 🎨 Yellow + Teal: Mild green 🎨 Purple + Teal: Dark indigo blue This approach provides clear insights into spatial relationships between markers, enhancing co-expression analysis. ➡️ Compatibility with Pathology Review: Chromogenic stains align well with traditional H&E evaluation, making them easier for pathologists to interpret. However, mIHC has its limitations: ⚠️ Challenge with multiple markers: Visualizing more than two co-localized markers becomes increasingly complex, as overlapping dyes can obscure clear interpretation. Color deconvolution requirements: If the tissue has high endogenous pigment or uneven staining, distinguishing true signals can be difficult. When to Go with #mIF? mIF utilizes fluorophores for marker detection, providing flexibility and clarity in studying complex tissue environments. Here’s why it might be a better fit: ➡️ Individual channel analysis: Each marker is visualized in its unique channel, eliminating the need for color deconvolution. ➡️ Co-localization of multiple markers: mIF allows easy detection and analysis of interactions among three or more markers in the same tissue region through composite views. ➡️ Digital analysis readiness: mIF pairs seamlessly with quantitative image analysis platforms, enabling precise, automated data extraction. Still, mIF has its own challenges: ⚠️ Autofluorescence: Some tissues, such as skin or liver, may produce background fluorescence, complicating signal interpretation. ⚠️ Fading signals: Fluorophores can photobleach, requiring careful imaging and handling protocols. ⚠️ Morphology visibility: While mIF offers detailed molecular insights, it may not always capture tissue morphology as clearly as chromogenic stains.
