PREMSTEM is a collective of world leading clinicians, researchers, stakeholder advocacy groups and an industrial partner with well-established experience in neonatology and drug development. We are developing a new regenerative stem cell therapy to repair the brain injury caused by premature birth, also known as encephalopathy of prematurity (EOP). World Health Day kicks off a year-long campaign on maternal and newborn health - a topic that is close to our heart in the PREMSTEM team! The campaign called 'Healthy beginnings, hopeful futures' urges governments and the health community to ramp up efforts to end preventable maternal and newborn deaths, and to prioritise women’s longer-term health and wellbeing. PREMSTEM partner GFCNI - Global Foundation for the Care of Newborn Infants connects families, healthcare professionals and researchers in the joint goal of advancing the health and quality of care for newborns and their families. Bobbi Fleiss and PhD candidate Victor Mondal from the Perinatal Brain Injury Lab at RMIT University are attending the PSANZ Congress this week in Brisbane, Australia. Bobbi presented two posters on behalf of her student, Md Munnaf Hossen. Victor presented 'Innovative Hydrogel Therapy: A Novel Approach for Neonatal Stroke Treatment'. Neonatal stroke occurs within the first 28 days of life. It can lead to long-term medical conditions such as cerebral palsy and epilepsy. Victor and the team developed a rat model of neonatal stroke and determined whether a functionalised hydrogel can repair injury and improve function. They found the model to produce a motor cortex lesion and functional deficits that is reduced by the functionalised hydrogel effectively. Lotte Smeets is a PhD student at the MERLN Institute and the Department of Paediatrics at Maastricht University where she is supervised by Sabine van Rijt, Tim Wolfs and Daan Ophelders. Together they are tracing the human mesenchymal stem cells that we use in PREMSTEM and unravelling their behaviour after administration in preclinical models of preterm birth. Thank you to Lotte for this image of nanoparticles and the nanoparticle-labelled MSCs. Lotte says that a major challenge in the clinical translation of MSC-based therapies is a limited understanding of how MSCs are distributed in the body. Tracing methods that can quantify the distribution of MSCs are needed to guide a more successful clinical translation of MSC-based therapies. "We synthesise gold nanoparticles containing a mesoporous silica coating. MSCs are labelled with gold nanoparticles (red signal in the right image) and are administered in vivo in order to determine the distribution of MSCs based on the gold core of the nanoparticles and their fluorescent coating," says Lotte. For Brain Awareness Week we're sharing the latest research highlights of one of our scientific publications. We create these summaries for people who would like to learn about our research without reading the full paper. 'Umbilical cord-mesenchymal stem cells induce a memory phenotype in CD4+ T cells' was co-authored by PREMSTEM researchers Ezgi Sengun, Tim Wolfs, Valéry van Bruggen, Daan Ophelders, Irma Joosten and Renate van der Molen. The goal of the study was to create a cell culture model to observe the long-term effects of human mesenchymal stem cells (H-MSCs) on activated T cells. The aim was to better understand how H-MSCs can influence the immune system and help manage inflammation, especially in vulnerable populations like preterm babies. The results suggest that stem cells from umbilical cord tissue or blood could help to control inflammation and immune responses, possibly offering a way to improve the body’s healing processes.
